  • The Brust-Schiffin synthesis of Organothiolate Monolayer Protected Gold clusters (Gold MPCs) created a new class of gold nanoparticles. These nanoparticles have the characteristics of unique stability, wide variety of facile surface functionality, size dependent capacitance, luminescence, and spectroscopic properties, and synthetic control over particle size. This represented an advance on the previous class of gold nanoparticles, but because of the inert nature of the new gold nanoparticles, the applications of the particles were limited. This invention describes the first method to overcome the inert property of the particles for the use of labeling a protein. It allows specific, discrete, and rigid affixment of the gold cluster on proteins or thiolated oligonucleotides, such as the single chain Fv antibody (scFv). Furthermore, this provides the necessary tool for use of electron microscopy for high resolution protein structure determination.
