Minor was born in Memphis, Tennessee but grew up in Flint, Michigan. He started doing stand-up and writing/performing with sketch groups in Detroit before joining the Second City Detroit mainstage. He is the only performer to have belonged to all three of the Second City's touring companies.
Minor was a featured player and staff writer on Saturday Night Live for one season, from October 2000 through the season finale in May 2001. He was let go from the show along with castmate Chris Parnell over the summer break, before the start of the 2001–2002 season because Lorne Michaels wanted to hire four new cast members, but did not want to go over budget.
